Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. Callus induction and plant regeneration of tomato (Lycopersicon esculentum cv. IPA 5) via anther culture Ciência Rural

Different growth regulators combinations were tested on the production of anther callus in tomato cultivar IPA 5. Calli were induced on media supplemented with 1.0mgL-1 gibberellic acid (GA3), 0.05mgL-1 alpha-naphthaleneacetic acid (NAA) plus 0.1mgL-1 6-benzylaminopurine (BAP), or with 1.0mgL-1 BAP plus 1.0mgL-1 NAA. The medium containing 1.0mgL-1 BAP and 1.0mgL-1 NAA produced the highest calli frequency, and promoted plant regeneration by indirect organogenesis, when calli were transferred to 0.01mgL-1 BAP and 0.001mgL-1 NAA. ABSTRACT The aim of this study was to evaluate different concentrations of two cytokinins in the in vitro propagation of the medicinal plant Hyptis marrubioides Epl. and to analyse the volatile fraction of the compounds. Nodal segments of plants were inoculated onto Murashige and Skoog (MS) solid medium with different concentrations of benzylaminopurine (BAP) or tidiazuron (TDZ) under a 16/8 h light/dark cycle at 25 ± 2 °C. After 45 days growth, the plants were evaluated for the number and length of the shoots and for shoot dry matter. The volatile constituents were analysed by headspace-GC/MS. ABSTRACT The aim of this study was to evaluate the effect of salicylic acid (SA) application on pre-transplant acclimatization of native populations of tomato. SA is a growth regulator that modifies plant growth and development by inducing changes in cell processes, physiology and morphology. Five populations of native tomato were sown in polystyrene trays. Peat moss was used as substrate and plants were maintained at field capacity continuously. This work focuses on the evaluation of flowering and vegetative growth of olive tree submitted to pruning and paclobutrazol application under field conditions with low temperatures during the winter. Two-years-old olive plant variety Grappolo 575, were submitted to light pruning, removing apical dominance, before treatments application. The treatments were organized in a 4×2 factorial scheme, respective to four PBZ concentrations tested: 0, 200, 400 and 800 mg L-1 of PBZ and two plants groups, with and without pruning, in randomized block with four replications. The biotechnological techniques may help solve many problems of guava culture, such as the high perishability of fruits. Somatic embryogenesis can generate highly multiplicative cell cultures and with high regenerative potential, serving as basis for genetic transformation. The aim of this work was to obtain somatic embryogenesis of guava (Psidium guajava L.) cv. Paluma. Immature seeds were used, and they were inoculated in MS environment containing 400 mg L-1 of L-glutamine, 100 mg L-1 myo-inositol, 60 g L-1 sucrose, 100 mg L-1 ascorbic acid and supplemented with different types and concentrations of growth regulators.
